<p><strong>Abstract</strong></p>
<p>Gynura procumbens (Lour) Merr., empirically, used to prevent cancer development and has been proven to be able to suppress lung cancer development. The aim of this research is to examine the potential of ethanolic extract of G. procumbens to suppress DMBA-induced breast cancer development. Sprague Dawly Rats were used in this research and were grouped as indicated treatment. Ethanolic extract of G. procumbens was administered into 3 levels of doses, namely 250, 500, and 750 mg/kgBW. Tumor development was examined by palpation every week and terminated at week 16th after the end of DMBA treatment. The result showed that extract treatment at the dose of 250, 500, and 750 mg/kgBW reduced tumor incidence by 60%, 30 %, and 20 % respectively. The doses of 500 and 750 mg/kgBW exhibited strong suppression of tumor multiplicity, where as the dose of 250 performed less potential suppression. In conclusion, ethanolic extract of G. procumbens performs chemopreventive effect to suppress breast cancer development at the dose of 250 mg/kgBW.</p>

<p>Breast cancer is the most common cancer occurring in women after cervix cancer in Indonesia. Tumor metastasis is the major cause of mortality in breast cancer. For a tumor cell to metastasize effectively, it must induce angiogenesis. 17 β-estradiol has been shown to stimulate the proliferation and angiogenesis of breast cancer cells which express estrogen receptor (ER), T47D (human breast cancer cell line). In the present study Pentagamavunon-1 or PGV-1 [2,5-bis-(4'-hydroxy-3',5'-dimethylbenzylidene)- cyclopentanone], an analogue of curcumin [1,7-bis-(4'-hydroxy-3'-methoxyphenyl)-1,6-heptadiena-3,5-dion], were tested on their cytotoxicity and suppression effect on angiogenic factors (i.e. VEGF and COX-2) on the breast cancer cell lines (T47D) induced by 17 β-estradiol 10-8 M. The results showed that PGV-1 performed cytotoxicity effect againts T47D cells with IC50 values 3,16 µM. This was more potent than curcumin (IC50 = 19,05 µM). PGV-1 5 µM and curcumin 20 µM decrease VEGF and COX-2 expression. These results suggest both compounds possessed antiangio-genic potensial.</p>

<p><strong>Abstract</strong></p>
<p>Gynura procumbens (Lour.) Merr., also known as Sambung Nyawa, has traditionally used as antipyretic, hypotensive, hypoglycemic and anticancer. Screening for fenolik compounds was done to the chemoprevention potency of Sambung Nyawa leaves. In the present work, the leaves crude extract was subjected to silica gel column chromatography and then eluted with n-hexane containing increasing amounts of ethyl acetate. Thin layer chromatography was carried out to perform the profile of 28 fractions. Fraction XIX-XX was selected to represent the relatively polar fractions. The fraction then was used for cytotoxicity test based on MTT method against HeLa cell line. By logit analysis, it was known that the IC50 of the fraction was 119 µg/ml. The experiment of antiproliferative effect was continued by studying the cell proliferation kinetics and DNA morphology. Result showed that the fraction was able to cause cell cycle delay and apoptosis. It assumed that the fraction had the antiproliferative effect so that this study can be used as a reference for chemoprevention trial.</p>

<p><strong>Abstract</strong></p>
<p>Breast cancer is the most common cancer occurring in women. Pentagamavunon-0 (2,5-bis-(4&#8242;-hidroxy-3&#8242;-methoksy)-benzilidine cyclopentanone: PGV-0), is a curcumin (1,7-bis-(4&#8242;-hidroksi-3&#8242;-metoksifenil)-1,6-heptadiena-3,5-dion) analogue that have been proved for its anticancer activity. The aim of this research is to study the antiproliferative effect of PGV-0 on breast cancer cell line, T47D, tested by MTT method. Inhibition ability of this compound evaluated by BrdU incorporation assay. The result of this research shown PGV-0 more potential as cytotoxic agent compare to curcumin by IC50 11 µM and 22 µM respectively. PGV-0 inhibits the cell growth started at 5 µM, and curcumin started at 10 µM. The antiproliferative effect was related to the inhibition of S-phase progression.. The result conludes that PGV-0 more potent than curcumin as antiproliferative agent on breast cancer cell line.</p>

<p>Pentagamavunon-0 or PGV-O (2,5-bis-(4&#8242;-hidroxy-3&#8242; methoxy) benzilidinecyclopentanone) is recognized as curcumin (1,7-bis-(4&#8242;-hidroxy-3&#8242;-metoksyphenyl)-1,6-heptadiena-3,5-dion) analogue that has been studied as anti-cancer. On breast cancer cell lines, curcumin shows apoptotic activity. The aim of this research is to study the potency of PGV-O to induce apoptosis on breast cancer cell line, T47D. Double staining methods with etidium bromide and acridine orange, DNA fragmentation assay, immunocytochemistry and western blot analyses were carried out to know the molecular mechanism of PGV-0 activity in apoptotic effect on T47D. The IC50 value of PGV-0 was 10,91 µM. PGV-0 induced the DNA laddering in DNA fragmentation assay and apoptotic activity was demonstrated by morphological analysis by using double staining methods. In accordance of the apoptotic effect , PGV-O increases the expression of BAX, decreases the expression BCl-2 and activate the caspase-3.</p>

<p>In previous study, the bark of ethanolic extract of Erythrina fusca exhibit cytotoxic activity on HeLa cell with IC50 of 11 µg/ml. The aims of this research are to examine cytotoxic activity of selected fraction of methanolic extract of the bark Erythrina fusca Lour and to study its inhibitory mechanism of the growing HeLa cells. Preperation of methanolic ekstract of the bark have been done by using maseration method, and the fractionation was carried out by using column chromatography. Fraction was grouped as similarity of the TCL profile, resulting in 10 fraction (A-J). Three fraction (F, G and I) were subjected to cytotoxic assay on HeLa cells. The higest cytotoxic activity of the fraction than tested for antiproliferative effect by measuring the doubling time and apoptotic effect by observing the treated cell morphology after staining with acrydine orange. The result showed the F fraction performed the best cytotoxic activity with IC50 of 76 µg/ml. In the concentration of 25 µg/ml the fraction prolonged the cell growth by twice compare to the control cells. . In the concentration of 50 µg/ml, the fraction induces apoptotic like body formation of the cells. These result suggest that the F fraction possess cycotoxic activity may by inducing cell cycle arrest and apoptosis.<br />
